TGF-β superfamily (TGF-β, BMP, activin) binds type-II/I Ser/Thr-kinase receptors; phosphorylates R-SMADs which partner with Co-SMAD4 → nuclear target genes. Roles: fibrosis, EMT, immunosuppression.
